Enzymes: describe question

Describe4 marksStandard

A student investigates the effect of pH on the activity of amylase. Two buffers of pH 7 and pH 9 are each mixed with a starch solution and the same volume of amylase. At regular intervals, a sample is removed and immediately mixed with iodine solution. The absorbance of each sample is measured at 620 nm using a colorimeter. The results are shown in the table.

Time / sAbsorbance at pH 7Absorbance at pH 9
00.850.85
300.600.72
600.350.60
900.150.50
1200.050.42

Describe how a colorimeter can be used in this investigation and how the results in the table can be used to compare the rates of reaction at the two pH values.

How this question is marked

4 marks, so the mark scheme is looking for about 4 separate creditworthy points. Making the same point twice in different words earns it once.

Describe means: Say what happens or what something is like, in order and with detail. You are not being asked why. If there is data, quote it: name the trend, then give figures with units at the points where it changes.
