Analytical techniques: describe question

Describe5 marksStandard

A student separates a mixture of amino acids using paper chromatography. The stationary phase is a sheet of cellulose paper, and the mobile phase is a non-polar solvent. The student marks a baseline in pencil near the bottom of the paper, applies spots of the mixture and of known amino acids, and places the paper in a tank containing the solvent. After the solvent has risen, the paper is dried and the amino acid spots are located.

Describe the meaning of the following terms, and state how the student would use them in this experiment:

(a) stationary phase (b) mobile phase (c) solvent front (d) baseline (e) Rf value

How this question is marked

5 marks, so the mark scheme is looking for about 5 separate creditworthy points. Making the same point twice in different words earns it once.

Describe means: Say what happens or what something is like, in order and with detail. You are not being asked why. If there is data, quote it: name the trend, then give figures with units at the points where it changes.
