Enzymes: compare question

Compare7 marksChallenge

Two enzymes, lactase (from E. coli) and cellulase (from Trichoderma reesei), catalyse the hydrolysis of different substrates. Their kinetic parameters are shown in the table.

EnzymeSubstrateKm (mM)Vmax (μmol min⁻¹ mg⁻¹)
Lactaselactose0.5120
Cellulasecellulose2.580

Compare the affinities of these two enzymes for their respective substrates. Explain the relationship between Km and enzyme–substrate affinity. Predict how the addition of a competitive inhibitor would affect the measured Km and Vmax for lactase, and justify your prediction.

How this question is marked

7 marks, so the mark scheme is looking for about 7 separate creditworthy points. Making the same point twice in different words earns it once.

Compare means: Give similarities or differences between two things, and say which side each point belongs to. Write each point as a pair, for example: A is X, whereas B is Y. Two separate descriptions rarely earn the marks.
